【摘要】:Thistestmethodisofprincipalvalueinminimizingthenumberofanimalsrequiredtoestimatetheacuteoraltoxicity(LD50).Thistestmethodisinappropriateformaterialstypicallyproducingdeath2ormoredaysafteradministrationofthetestcompoundunlesstheobservationtimebetweendosagesisincreased.Thistestmethodcanbesuccessfullyapplied,however,formaterialsproducingonlyanoccasionaldeath2ormoredaysafteradministration.TheLD50isvaluableasameasureoftherelativeacutetoxicityofamaterialandcanbeusedtomakeanestimateofpotentialhazardtohumanswhenpesticides,otherchemicals,ormixturesareingested.Thistestmethodallowsforobservationofsignsoftoxicityinadditiontomortality.Thisinformationcanbeusefulinplanningadditionaltoxicitytesting.1.1Thistestmethoddeterminesthelethality(LD50value)andsignsofacutetoxicityfromamaterialusingalimitednumberofrats.Thetechniqueusedinthistestmethodisreferredtoasthe"Up-and-DownProcedure."ThistestmethodisanalternativetotheclassicalLD50testandisapplicabletobothliquidsandsolids.1.2Thistestmethodisnotrecommendedfortestmaterialswhichtypicallyproducedeathsbeyond2dayspostdosing.1.3Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.
